PRODUCT DESCRIPTION
The Super Hyb Kit is a complete hybridization system that provides exceptionally high signal intensity and low background on spot-free northern, Southern and dot-blot hybridizations. This system allows detection of rare mRNA in total RNA and can eliminate the need to isolate poly A+ RNA. The Kit provides fast and simple procedures and works well with radioactive or non-radioactive detection methods.

The Super Hyb Kit includes two solutions. The High Efficiency Hybridization System (without or with formamide, HS 114 and HS 114F, respectively) increases hybridization efficiency by blocking nonspecific binding of probes and enhancing hybridization signal intensity. The Washing & Pre-Hyb Solution (cat. no. WP 117) allows both pre- and post-hybridization washes to be performed at room or elevated temperature.

APPLICATION
Perform pre-hybridization, hybridization and post-hybridization procedures according to the protocol presented in the product application sheet for the High Efficiency Hybridization System. The High Efficiency Hybridization System product application sheet with a detailed hybridization protocol accompanies this Super Hyb Kit.

Depending upon membrane area and the size of bags, tubes or trays used for hybridization and washing procedures, hybridization of most membranes requires 5 - 10 ml of HS 114 or HS 114F while washing steps require a total of 40 - 50 ml of the WP 117 (provided as a 10X concentrate; dilute to a 1X working solution). The 1X working solution is stable for two weeks after dilution. The Super Hyb Kit may be used with nitrocellulose or a variety of charged or non-charged nylon membranes. The Super Hyb Kit is available in small and large volumes, and reagents may also be purchased individually.
